Description

Thermolabile Exonuclease I catalyzes the removal of nucleotides from single-stranded DNA (ssDNA) in the 3′ to 5′ direction.

Thermolabile ExoI can be heat inactivated quickly to speed up downstream applications. This data demonstrates that Thermolabile ExoI can be heat inactivated at 80°C for 1 min, 65°C for 4 min or 60°C for 15 min in both NEBuffer 3.1 as well as Standard Taq Reaction Buffer.

Unit Definition

One unit of Thermolabile Exonuclease I is defined as the amount of enzyme that will catalyze the release of 2 nmol of acid-soluble nucleotide in a total reaction volume of 100 μl in 6 minutes at 37°C in 50 mM Tris-HCl (pH 7.9), 100 mM NaCl, 10 mM MgCl2 and 100 μg/ml BSA with 0.17 mg/ml single-stranded [3H]-E. coli DNA.

Product Notes

  1. This enzyme can be heat inactivated at 80°C for 1 minute, 65°C for 5 minutes or 60°C for 15 minutes.
  2. Thermolabile ExoI is active in the presence of 25 mM to 200 mM NaCl.
  3. If the amount of ssDNA to be degraded needs to be increased, we recommend adding more enzyme instead of extending the reaction time.
